Mesenchymal stem cells (MSCs) derived from the amniotic membrane—the innermost layer of the placenta—constitute a developmentally young, immunologically permissive population with distinctive regenerative properties. The amniotic membrane itself has been used clinically for centuries in wound healing and ocular repair; extraction of its cellular component yields MSCs that retain the tissue's inherent anti-inflammatory and pro-angiogenic characteristics. These placental MSCs differentiate efficiently into bone, cartilage, and fat lineages and secrete a rich cocktail of cytokines and growth factors documented to suppress inflammatory responses. Sourcing is non-invasive relative to bone marrow harvest, and placental tissue would otherwise be discarded after birth, raising fewer ethical concerns than embryonic sources. The cells expand readily in culture and are readily cryopreserved, enabling off-the-shelf banking. Allogeneic placental MSCs have entered clinical trials for acute graft-versus-host disease, inflammatory bowel disease, and joint cartilage repair, with safety signals emerging from early-phase studies. Their mechanisms appear to centre on immune regulation and tissue-protective signalling rather than engraftment as structural elements. Long-term efficacy and optimal patient selection criteria are still being refined. The relative abundance of placental material and established isolation protocols position these cells as a scalable therapeutic modality, though standardisation across different producers remains a regulatory and quality consideration.
