Status: Legal supplement everywhere. One of the most-studied molecules in the longevity field — and one of its biggest cautionary tales.

Resveratrol is what a longevity hype cycle looks like after 20 years: more than 21,000 indexed papers and roughly 9,700 human records, yet no convincing human benefit at supplement doses. The famous sirtuin-activation story weakened under replication attempts, a major industry programme was abandoned, and human trials keep returning null or marginal results — partly because oral bioavailability is extremely poor.

What it is

Resveratrol is a polyphenol from grape skin and Japanese knotweed that shot to fame in the 2000s as the proposed explanation for the “French paradox” and as a sirtuin activator that extended lifespan in some model organisms.

What the research actually shows

Human trials across metabolic health, cardiovascular markers and inflammation have produced inconsistent, mostly small or null effects. The molecule is metabolised within minutes of ingestion, so plasma levels of the parent compound stay far below anything used in cell studies. The original sirtuin-activation biochemistry was later shown to depend partly on an assay artefact, and follow-up animal work found lifespan extension only in specific contexts such as high-fat diets.

Why it still matters

Resveratrol is the reference case for reading longevity headlines: spectacular mechanism press coverage, billions in market value, and two decades later no demonstrated human aging outcome. Any new molecule marketed on cell-culture sirtuin data should be weighed against this precedent.
