Masayo Takahashi

First iPS-cell clinical transplant · b. 1961

Masayo Takahashi led the first iPS cell clinical transplantation in 2014, implanting iPS-derived retinal pigment epithelial (RPE) cells into a patient with age-related macular degeneration (AMD). This landmark procedure demonstrated that reprogrammed cells could be expanded in culture, differentiated into a functional cell type, and safely introduced into a human patient. Takahashi's meticulous characterisation of the iPSC-derived RPE—assessing gene expression, protein production, and functional properties—established a template for translational validation of stem cell products. The procedure itself showed remarkable technical precision: injection of a single layer of differentiated cells into a precisely defined anatomical location, with post-operative imaging confirming graft survival and integration. Although the trial was subsequently expanded and then paused after one patient developed a potential adverse event (later attributed to infection risk rather than cell oncogenicity), Takahashi's work validated the feasibility of iPS cell therapy and provided crucial safety data. His contributions include not only the clinical execution but also the regulatory strategy and manufacturing process development necessary to move a cell therapy from laboratory prototype to clinical candidate. Takahashi's pioneering work in regenerative ophthalmology has influenced subsequent clinical trial designs for RPE replacement and cell therapy delivery generally, establishing scientific and operational principles that continue to guide translational stem cell medicine.
